Sequence Design & Optimization:
Anemocyte built its platform offering mRNA construct design and optimization services. These can include codon optimization (to maximize protein expression in the target species or cell system), modification of untranslated regions (UTRs) to improve stability and translational efficiency, and engineering of the poly(A) tail. Alternatively, the sequence design can originate from the Customer. In this case Anemocyte is able to produce mRNA using of sequences designed by them based on the proteins they wish to express. -
Production of DNA template for in vitro transcription.
Anemocyte has a strong expertise in plasmid DNA production. Hence, Anemocyte can offer plasmid production and linearization services as part of our mRNA synthesis platform. Otherwise, Anemocyte can produce mRNA starting from DNA template provided by our customers. - mRNA Modifications:
Anemocyte offer various modifications to improve stability, reduce immunogenicity, and increase translational efficiency of mRNA. 5' Capping: Essential for translation initiation and mRNA protection. Options include co-transcriptional capping or post-transcriptional enzymatic capping using our proprietary patent-pending technology. Incorporation of Modified Nucleosides: The use of nucleosides like pseudouridine (Ψ), N1-methylpseudouridine (m1Ψ), or 5-methylcytidine (m5C) helps to evade the host's innate immune response and can increase mRNA persistence and translation. PolyA modification: Precisely tailored poly(A) tails are included in the plasmid DNA template to fine-tune mRNA's performance. - Rigorous Purification:
Anemocyte has developed different purification strategies to ensure exceptionally pure mRNA. Our purification techniques typically include chromatography and tangential flow filtration to remove contaminants such as the DNA template, enzymes, free nucleotides, proteins, and, crucially, double-stranded RNA (dsRNA), for optimal safety and efficacy. - Comprehensive Quality Control:
Anemocyte performs in house nnumerous analytical tests to ensure the identity, purity, integrity, concentration, and safety of the produced mRNA. Common tests include: spectrophotometry (A260/A280), gel electrophoresis (to verify size and integrity), analytical HPLC (for purity), and, if applicable, in vitro potency or expression assays. - Scalability to Meet different demands.
